چکیده

Various reports (l-5) have demonstrated that streptomycin inhibits the metabolic activities of the non-proliferating cells of susceptible strains of bacteria. This paper is an attempt to trace the mode of action of streptomycin with the object of determining what reactions vital to the cell are interfered with. Study of the mode of action of the antibiotics of therapeutic value should eventualIy provide an explanation for their ability to enter the animal body and kill susceptible bacteria without killing the tissue cells. The first step toward this goal is to determine which reactions of the bacterium are susceptible to streptomycin inhibition at concentrations of this agent which seem to be effective in practice. In order to investigate this problem, we began with the observations of Geiger (3), who demonstrated that, with a particular strain of Escherichiu IX& there was a marked stimulation of serine oxidation by the previous oxidation of fumarate. When streptomycin was present (at levels of 5 to 26 y per ml.) during the oxidation of fumarate, this stimulation was prevented. Geiger concluded that streptomycin prevents the action of some intermediate, formed during the oxidation of fumarate and a variety of other carbohydrate substances, which is necessary for the oxidation of serine. One could also interpret the data reported by Geiger as indicating that it was the jorwmtion of the intermediate which was interfered with by streptomycin. Using these observations as a starting point, we have been able to trace the mode of action of streptomycin somewhat further.

Antibiotic Supplements Affect Electrophysiological Properties and Excitability of Rat Hippocampal Pyramidal Neurons in Primary Culture

Introduction: Antibiotic supplements are regularly used in neuronal culture media to control contamination however, they can interfere with the neuronal excitability and affect electrophysiological properties. Therefore, in this study, the effect of penicillin/streptomycin supplements on the spontaneous electrophysiological activity of hippocampal pyramidal neurons was examined. Methods: Electr...

The field monitoring of influential biodeteriogenic agents on the historic rock surfaces in Persepolis-UNESCO World Heritage Site

Cultural heritage has always been the focus of many civilizations and therefore, it needs to be preserved for future generations. From prehistoric times, when grandeur and beauty were the aims of architecture, stone was the most widely used durable material. Biodeterioration of the stone monuments, one of the most important causes for the loss of the cultural heritage, is defined as any undesir...

Mechanism-Based Studies of the Active Site-Directed Inhibition and Activation of Enzyme Transketolase

Derivatives of phenyl-keto butenoic acids have been reported to be inhibitors of pyruvate decarboxylase, (PDC). The inhibition of transketolase, a thiamine requiring enzyme such as PDF, by meta nitrophenyl derivative of 2-oxo-3-butenoic acid (MNPB) is reported here. These studies indicate that the inhibitor binds to the enzyme at the active site. A two-step inhibition was observed, first th...

Aminoglycoside uptake and mode of action-with special reference to streptomycin and gentamicin. II. Effects of aminoglycosides on cells.

Introduction Target theory and the lethal event. In considering the mode of action of aminoglycosides like streptomycin and gentamicin on bacterial cells, it is first worthwhile to describe exactly what is meant when one describes them as bactericidal. According to the target theory of drug action proposed by Paul Ehrlich (see Setlow & Pollard, 1962 for discussion), survival represents the tota...
